Overview

Chorionic gonadotropin subunit beta (hCGβ) is a glycoprotein hormone primarily produced by the placenta to maintain the corpus luteum and progesterone production during early pregnancy (UniProt P01233). Beyond its reproductive role, hCGβ is frequently expressed by various epithelial cancers, including colorectal, lung, and pancreatic malignancies, where it functions as an autocrine and paracrine growth factor (PMID: 32824110). In these settings, hCGβ promotes tumor cell proliferation, inhibits apoptosis, and facilitates immune evasion by modulating the tumor microenvironment. As a vaccine antigen, hCGβ is utilized to stimulate the production of specific antibodies that neutralize the hormone, thereby disrupting its oncogenic signaling or preventing embryo implantation in contraceptive applications (PMID: 16153770). Clinical development focuses on overcoming immune tolerance to this self-antigen by using recombinant proteins or peptide conjugates, often targeting the unique C-terminal peptide (CTP) to avoid cross-reactivity with luteinizing hormone (NCT00404742).

Mechanism of action

Induction of active immunity through the production of neutralizing antibodies that bind to circulating or membrane-bound hCG beta subunit, thereby blocking its interaction with the LH/hCG receptor and inhibiting its pro-tumorigenic or pro-gestational effects.
