Overview

Chorionic somatomammotropin hormone 2 (CSH2) is a placental hormone produced exclusively during pregnancy and is a member of the somatotropin/prolactin family of hormones. CSH2 is primarily expressed in the placenta and is involved in stimulating lactation, promoting fetal growth, and modulating maternal and fetal metabolism during gestation. Unlike growth hormone (GH), it does not interact with the growth hormone receptor (GHR) but instead activates the prolactin receptor (PRLR), influencing maternal metabolic adaptation to pregnancy and supporting fetal development. CSH2 shares high sequence identity with related placental lactogens, and differential expression is developmentally regulated. Genetic variants or dysregulation of CSH2 have been associated with gestational trophoblastic diseases, such as choriocarcinoma and epithelioid trophoblastic tumor.
