Overview

Chromatin licensing and DNA replication factor 1 (CDT1) is a key protein encoded by the *CDT1* gene that regulates DNA replication. It is an essential licensing factor for the assembly of pre-replication complexes at replication origins, ensuring that DNA replication occurs only once per cell cycle. CDT1 cooperates with CDC6 and the origin recognition complex during the G1 phase; its activity is negatively regulated after initiation by inhibitory proteins such as Geminin and by targeted degradation. Dysregulation of CDT1 is associated with genomic instability, cancer, and developmental syndromes such as Meier-Gorlin syndrome.
