Overview

Chromatin-remodeling ATPase INO80 (INO80) is the catalytic subunit of the multisubunit INO80 complex, a member of the SNF2 family of ATP-dependent chromatin remodelers[1][2][4]. INO80 features a Swi2/Snf2-type ATPase domain and unique insertion regions that organize the recruitment and assembly of other complex subunits[1][2]. The INO80 complex catalyzes chromatin remodeling through ATP-dependent nucleosome sliding and histone variant exchange, acting as a processive 'motor' that repositions nucleosomes and modulates DNA accessibility for transcription, repair, and replication[2][4]. It is essential for genomic DNA homeostasis, regulating promoter architecture, influencing gene expression, repairing double-strand DNA breaks, and facilitating replication fork progression[2][4]. Dysregulation of INO80 is implicated in cancer and other genomic stability disorders, but it is not yet a direct therapeutic target; efforts to modulate its function remain mostly preclinical[2].
