Overview

Chromobox protein homolog 1 (CBX1), also known as HP1β, is a highly conserved non-histone chromosomal protein and a key regulator of heterochromatin structure and function[1][2][3][5]. CBX1 is enriched in heterochromatin and is essential for gene repression through chromatin compaction, primarily by binding to histone H3 trimethylated at lysine 9 (H3K9me3) via its N-terminal chromodomain[1][2][3][5]. It also features a C-terminal chromoshadow domain that enables homodimerization and interactions with other chromatin-associated proteins[1][2][3]. CBX1 mediates various critical processes in the cell, including the maintenance of genome integrity, telomere homeostasis, and the cellular response to DNA damage. It is a central factor in the epigenetic silencing of genes and is implicated in diseases such as cancer due to its role in transcriptional repression and chromatin dynamics[5][3].
