Overview

Chromobox protein homolog 2 (CBX2) is a member of the Polycomb group (PcG) family, functioning as a chromatin regulatory protein within the canonical Polycomb repressive complex 1 (cPRC1). CBX2 is essential for the compaction of chromatin and the formation of phase-separated nuclear condensates, which are critical for stable transcriptional repression of lineage-inappropriate genes in development and stem cell differentiation[1][2]. It acts as a scaffold in Polycomb nuclear condensates, organizing PcG-bound chromatin domains and mediating long-range chromatin interactions, primarily via its intrinsically disordered regions and basic amino acid-rich segments. CBX2 expression is tightly regulated during development, being upregulated during differentiation events such as spermatogenesis, and is critical for repressing genes active in stem cells and for the maintenance of certain adult stem cell lineages. Loss of CBX2 function in mice leads to severe developmental defects, including axial patterning abnormalities, bone growth defects, and problems with sex determination, highlighting its crucial role in development and tissue homeostasis[1]. In cancer biology, CBX2 has been implicated as a potential oncogenic protein due to its capacity to alter chromatin compaction and repress tumor suppressor genes; however, it is not a direct target of approved therapeutic drugs as of now[1][2].
