Overview

Chromodomain-helicase-DNA-binding protein 4 (CHD4) is an ATP-dependent chromatin-remodeling enzyme of the SNF2/RAD54 helicase family and is a core component of the nucleosome remodeling and deacetylase (NuRD) complex[2][1][5]. It binds and distorts nucleosomal DNA and plays a key role in epigenetic transcriptional repression, DNA damage repair (notably via homologous recombination), and arrest of transcription in response to DNA damage[2][5][1]. CHD4 has been implicated in neurogenesis, and its dysfunction or mutation is associated with several cancers, developmental syndromes (notably Sifrim-Hitz-Weiss syndrome), and is a known autoantigen in dermatomyositis[2][5][4]. Its targeting is of interest for modulating gene expression and DNA repair pathways in disease contexts[1][2].
