Overview

Chromodomain Y-like protein (CDYL) is a transcription corepressor ubiquitously expressed in humans, distinct from its testis-specific paralogs on the Y chromosome[4]. CDYL possesses an N-terminal chromodomain and a C-terminal enoyl-coenzyme A hydratase (ECH) catalytic domain, enabling it to act as both a chromatin-binding reader and a putative enzyme[2]. It is part of multimeric repressive chromatin complexes and is required for faithful transmission and restoration of repressive histone marks such as H3K9me2/3 and H3K27me2/3 during DNA replication, playing an essential role in preserving epigenetic information and cell identity[1][5]. CDYL bridges chromatin assembly factor 1 (CAF-1) and components of the MCM replicative helicase complex to facilitate the inheritance of repressive histone modifications and genome stability. It also recruits histone-modifying enzymes including G9a, SETDB1, and EZH2 to replication forks. Loss of CDYL function impairs DNA damage repair (particularly homologous recombination), S phase progression, and can sensitize cells to genotoxic stress[1]. CDYL is genetically associated with infertility, some cancers, and may be involved in other disease processes, but is not currently a direct therapeutic target for drugs[4][2].
