Chromosome 1 open reading frame 159 (C1orf159) encodes a largely uncharacterized, evolutionarily conserved, proline- and arginine-rich protein whose main feature is a DUF4501 domain of unknown function. The protein is predicted to be a single-pass, membrane-associated protein with a signal peptide and a transmembrane domain. Its main isoform is 380 amino acids (40.38 kDa) but alternative splicing produces additional isoforms. C1orf159 is thought to act as a scaffold/adaptor protein, possibly involving protein-protein interactions and membrane localization, and may play a role in brain development and protein ubiquitination. While its molecular function remains unclear, it has prognostic significance in several cancers. High expression correlates with poor prognosis in renal and liver cancer and better prognosis in urothelial cancer. There are no known drugs targeting C1orf159 and no established mechanism of action for therapeutics. The gene is located at chromosome 1p36.33.
