Chromosome 1 open reading frame 167 (C1orf167) is an uncharacterized protein encoded by the C1orf167 gene on chromosome 1p36.22 in humans[1][7]. The protein is 1468 amino acids in length, with a molecular weight of approximately 160–162 kDa; it is predicted to be localized mainly to the cell nucleus and is subject to post-translational modifications such as phosphorylation, glycosylation, SUMOylation, and glycation[1]. C1orf167 is expressed at higher levels in the testis, larynx, blood, placenta, and prostate[1]. Although the protein has a conserved domain of unknown function and is rich in alpha-helical structures, its precise function remains uncharacterized[1][2]. Clinically, increased expression has been observed in certain cancers (e.g., leukemia, head, neck, lung), as well as in infectious diseases like Chlamydia pneumoniae infection and tuberculosis, and it may be implicated in coronary artery disease[1][2][7]. There are no established drugs targeting this protein, and it is not currently considered a validated therapeutic target.
