Overview

Chromosome 1 open reading frame 21 (C1orf21) encodes an intracellular protein, most highly expressed in the skin, lung, and prostate, but also found at low to moderate levels in most tissues[1][11][2]. The protein is 121 amino acids (about 18.7 kDa) and contains Domain of Unknown Function 4612 (DUF4612)[1][12]. C1orf21 shows subcellular localization primarily in the nucleus and cytoplasm and is predicted to interact with various proteins involved in chromatin assembly, transcriptional regulation, inflammation, and developmental processes, including Cabin1, CEP162, CD97, NKX2-8, and SEMA3C[1]. Although its precise molecular function is still unclear, C1orf21 is linked to cell growth and reproduction, and several studies have found associations with cancer, particularly in relation to cell proliferation and keratinocyte regulation[1][10][11]. There are no known isoforms, paralogs, drug interactions, biomarker roles, or safety concerns documented in the current literature[1][3][6][10]. The gene is well-conserved evolutionarily, with orthologs found in many vertebrates and invertebrates[1].
