Chromosome 1 open reading frame 56 (C1orf56), also named Protein MENT, is a human protein coded by the C1orf56 gene on chromosome 1. The protein is implicated as a proto-oncogene whose promoter is methylated and repressed by DNA methyltransferase 3B (DNMT3B); in certain lymphomas, a catalytically inactive DNMT3B isoform leads to loss of promoter methylation, derepressing C1orf56 expression. C1orf56 plays a role in cellular proliferation and is described as an onconcogenic modifier with a potential contribution to tumor suppressor activity of DNMT3B. There is a reported link between C1orf56 gene variants and corneal dystrophy (posterior polymorphous type). The molecular function remains incompletely defined and it is not established as a receptor, enzyme, transporter, or transcription factor; instead, it is categorized as an "other" protein with proto-oncogenic and proliferation-associated activities. No drugs, biomarkers, or safety issues are currently documented for clinical targeting of C1orf56.
