C1orf74 encodes a protein of 269 amino acids containing the DUF4504 domain, expressed broadly across human tissues. It is upregulated in lung adenocarcinoma and high expression correlates with unfavorable prognosis. In LUAD cell models, knockdown of C1orf74 inhibits cell proliferation, migration, and invasion, induces cell cycle arrest, and increases autophagy. Mechanistically, C1orf74 positively regulates EGFR/AKT/mTORC1 signaling, affects cyclin D1 expression, and may act as an oncogene, suggesting its potential as a novel cancer biomarker and therapeutic target. Its locus is associated with schizophrenia risk and may interact genetically with the IRF6 gene, linked to congenital diseases. No direct drug interactions or mechanisms of action for approved medicines have been described, and its structural or molecular family is classified as "other" due to an unknown function apart from containing DUF4504.
