Overview

Chromosome 1 open reading frame 94 (C1orf94) is a human protein of unknown function, coded by the C1orf94 gene located at chromosome 1p34.3[1][8][10]. It is a protein-coding gene that has been identified as an “uncharacterized protein” in major protein and genetic databases[5][8][9]. C1orf94 is known to encode at least two isoforms, with the longer being 598 amino acids, and is predicted to localize primarily to the cell nucleus with high testis tissue expression and slight brain expression[1][6][10][11]. The protein contains a conserved DUF4688 domain and several PEST motifs, suggesting it may have a short intracellular half-life[1]. While there is evidence for possible physical interactions with proteins involved in chromatin binding and cell signaling, such as ATXN1 and MMADHC, the precise biological function of C1orf94 remains uncharacterized[1]. Genome-wide studies have identified C1orf94 as an oncogenic open reading frame (OncoORF) potentially implicated in tumorigenesis—particularly in the context of colorectal cancer through its interaction network—but no direct functional studies or therapeutic targeting have been reported[1]. The available data do not support C1orf94 as a classical therapeutic target (such as a receptor, enzyme, transporter, etc.), and it currently lacks characterized biological functions, known interacting drugs, or recognized roles as a biomarker or safety concern[1][2][5][8][10].
