Chromosome 16 open reading frame 46 (C16orf46) is an uncharacterized protein encoded by the C16orf46 gene in humans and is most commonly referred to by its gene symbol C16orf46 or the alias FLJ32702[1][3][5]. The protein consists of 417 amino acids, is predicted to be located in the nucleus, and shows alternating alpha helices and beta sheets in its secondary structure[1]. C16orf46 is broadly expressed at low to moderate levels in several tissues, most notably testis, thyroid, kidney, liver, prefrontal cortex, cerebellum, heart, and brain[1]. Despite its conservation across vertebrates, the specific biological function of C16orf46 remains unknown; current data do not classify it within any major protein family such as receptor, enzyme, transporter, or transcription factor[1][3]. There is evidence implicating C16orf46 as a potential prognostic biomarker for hepatocellular carcinoma (HCC), meaning altered gene expression may help predict patient outcomes, but the underlying molecular mechanisms have not been elucidated[3]. Elevated C16orf46 expression has also been observed in pancreatic adenocarcinoma and small-cell carcinoma tissues relative to controls[1]. There are no approved drugs known to directly interact with this protein, nor are there any characterized mechanisms of drug action targeting it. There are also currently no established safety concerns associated with C16orf46, primarily due to the lack of functional and therapeutic studies on this uncharacterized protein.
