Overview

Chromosome 16 open reading frame 54 (C16orf54) is a human protein-coding gene encoding a transmembrane protein with biased expression in hematopoietic and immune tissues, such as bone marrow, lymph node, plasma, and peripheral blood mononuclear cells[2][8]. Its functions are not yet fully elucidated, but C16orf54 is recognized as *Surface Antigen In Leukemia (SAIL)* and studied as a potential cell surface marker in certain cancers, particularly hematological malignancies[6][2]. Integrated pan-cancer expression analyses indicate heterogeneous regulation across tumor types: C16orf54 is downregulated in several cancers (e.g., breast, colon, lung), while upregulated in others (e.g., glioblastoma, renal carcinomas, leukemia), and its expression is linked to prognosis, immune infiltration, and stemness features across cancers[2][4]. C16orf54 demonstrates high accuracy in distinguishing normal from malignant tissues and is positively correlated with immune microenvironment scores, immune cell infiltration, chemokine and receptor signatures, and multiple immune signaling pathways (including cytokine-cytokine receptor interactions, T cell receptor signaling, and inflammatory pathways)[2][4]. While no drugs directly targeting C16orf54 have been described, its potential as a diagnostic, prognostic, and immunotherapy marker in oncology is under investigation[2][4].
