Chromosome 19 open reading frame 25 (C19orf25) encodes a small, poorly characterized protein belonging to the UPF0449 family, found only in bony vertebrates. It acts as a cytoplasmic protein that participates in the NRZ complex—an evolutionarily conserved multiprotein assembly involved in retrograde traffic from the Golgi apparatus to the endoplasmic reticulum, and potentially has a regulatory or scaffolding function linking vesicle tethering, cell cycle checkpoints, and autophagy. C19orf25 has been shown to physically interact with other trafficking proteins such as USE1 and STING, and its knockout impacts retrograde transport and STING signaling, a pathway relevant to innate immunity. C19orf25 is localized to a genomic region (19p13.3) associated with neurodevelopmental syndromes when deleted, but it is not currently recognized as a direct therapeutic target, nor are there known drugs acting on C19orf25 in the clinic.
