Overview

Chromosome 19 open reading frame 84 (C19orf84) is a protein-coding gene encoding a protein adapter involved in the silencing of transposable elements via piRNA-mediated DNA methylation in the mammalian germ line[1][3][5]. C19orf84 acts as a connector that enables effective cooperation between the piRNA pathway component PIWIL4 (also known as MIWI2 in mice), the co-factor SPOCD1, and DNA methylation machinery such as DNMT3C and DNMT3L, particularly during the development of male germ cells[1][3][4][5]. Through this role, C19orf84 ensures the maintenance of genomic stability by suppressing the mobilization of transposons, a vital process for fertility and genome integrity in mammals[1][3][5]. Loss of C19orf84 function disrupts these processes, leading to defective silencing of transposable elements and male infertility in mouse models, and may contribute to similar defects in humans[3][4]. There is no current evidence that C19orf84 is a direct therapeutic target, receptor, enzyme, or transporter; instead, it represents a specialized adapter protein that is essential for constitutional genome defense during spermatogenesis and is implicated in the biology of male infertility[3][4][5].
