Overview

Chromosome 2 open reading frame 72 (C2orf72) is a protein-coding gene in humans encoding an uncharacterized intracellular protein, also referred to as "uncharacterized protein C2orf72" or LOC257407. The precise molecular function of C2orf72 is currently unknown, but it is predicted—mainly from gene expression and computational analyses—to be involved in very-low-density lipoprotein (VLDL) particle assembly and in the regulation of cholesterol esterification. Expression of C2orf72 is seen predominantly in the liver, brain, placenta, small intestine, and colon, with the protein localized mainly to the cell membrane and cytoplasm, and possibly in the nucleus. Both estradiol and testosterone have been shown to upregulate C2orf72 expression, and its expression is reduced in the absence of hepatocyte nuclear factor 4 alpha (HNF4α). No established roles in disease, therapeutic targeting, or drug interaction are currently reported for C2orf72. It interacts with proteins NRas, KRas, and CD81, but the functional significance of these interactions is also uncharacterized.
