Overview

Chromosome 2 open reading frame 92 (C2orf92), also known as LINC01125, is a gene located on human chromosome 2 that has been variably annotated as encoding an uncharacterized protein and as a long non-coding RNA (lncRNA). The locus is predicted to be membrane-associated but, functionally, most studies characterize its major transcript as the lncRNA LINC01125, rather than a protein product. LINC01125 is implicated in the suppression of tumorigenic phenotypes in cancer cell models, acting by sponging microRNAs (e.g., miR-19b-3p) and regulating targets such as TNFAIP3, thus affecting cell proliferation, invasion, and migration. The expression of LINC01125 is altered in several cancers, and exosomal LINC01125 in serum is suggested as a noninvasive biomarker for NSCLC diagnosis and prognosis. Currently, there are no known drugs targeting C2orf92/LINC01125, and it is not established as a therapeutic target in the conventional sense (e.g., receptor, enzyme). There is inconsistency and lack of clarity regarding whether C2orf92 should be classified as a protein-coding gene, a lncRNA gene, or both, and the "uncharacterized protein" designation highlights the uncertainty and incomplete functional annotation.

Mechanism of action

For the lncRNA LINC01125: acts as a competing endogenous RNA (ceRNA) sponging miR-19b-3p to regulate TNFAIP3; also subject to regulation by m6A modification via METTL3
