Overview

Chromosome 4 open reading frame 46 (C4orf46), also known as renal cancer differentiation gene 1 (RCDG1), is a small, conserved protein of unknown function encoded by the C4orf46 gene on chromosome 4. The gene is expressed in multiple human tissues, but its molecular and cellular roles remain uncharacterized. Recent clinical research has shown that C4orf46/RCDG1 expression is significantly downregulated in renal cell carcinoma (RCC) tissue compared to adjacent normal tissue, with lower expression in clear cell RCCs and an inverse correlation with tumor grade, suggesting it may have a role in tumor differentiation or progression and could serve as a prognostic biomarker in renal cancer. There are no known drugs, mechanisms of action, or specific safety issues associated with targeting this protein to date, and it is not currently considered a therapeutic target.

Mechanism of action

None known (no drugs or biological effectors known to target this protein)

03

Biological functions

Unknown (no defined biochemical or molecular function in the literature as of current knowledge)Possible role in cell differentiation, particularly in the context of renal cell carcinoma, based on expression correlation
04

Disease associations

Cancer (specifically, reduced expression in renal cell carcinoma and association with disease grade)Other (no defined causality or mechanistic involvement in other diseases, but associated with some cancers in gene-disease panels)
05

Safety considerations

None reported (no therapeutic targeting documented, hence no documented safety concerns)
06

Interacting drugs

None known or reported in the literature
07

Biomarkers

Possible biomarker for prognosis in renal cell carcinoma (downregulation associated with higher Fuhrman grade and clear cell subtype of renal carcinoma)
