Overview

Chromosome 5 open reading frame 34 (C5orf34) encodes a conserved protein of 638 amino acids in humans, containing two domains of unknown function (DUF 4520 and DUF 4524) and a predicted polo-box domain associated with kinase activity[1]. Its expression is relatively low in most tissues but highest in stomach, intestine, testis, skeletal and heart muscle, and is also detected in dermal fibroblasts[1]. Despite its poorly understood biological function, structural predictions suggest involvement in kinase-related nuclear functions and regulation of cell proliferation[1]. C5orf34 is significantly overexpressed in various cancers and correlates with poor prognosis, likely through influence on the immune microenvironment, making it a candidate prognostic biomarker and a potential therapeutic target[2].
