Chromosome 8 open reading frame 88 (C8orf88) is a small human protein (117 amino acids) identified as a novel eIF4E-binding protein with homology to the canonical eIF4E-binding proteins (4E-BPs). C8orf88 is predominantly expressed in the testis, specifically in spermatocytes and early spermatids, suggesting a specialized role in the regulation of translation during spermatogenesis. Experimental data show that C8orf88 binds eIF4E via a conserved eIF4E-binding motif (4EBM) and negatively regulates global protein synthesis (translation) when overexpressed in cultured cells. Unlike canonical 4E-BPs, C8orf88 lacks most mTOR-regulated phosphorylation motifs, indicating it may be regulated by different mechanisms. While aberrant function of 4E-BP family members has been implicated in cancer, and C8orf88 is expressed in germ cells where germ cell tumors arise, its direct role in human disease remains to be established. There are currently no well-characterized drugs, mechanisms of action, or biomarkers associated with C8orf88, and it is not widely recognized as a therapeutic target.
