Chromosome alignment-maintaining phosphoprotein 1 (CHAMP1) is a zinc finger protein crucial for proper chromosome segregation during mitosis by facilitating the attachment of kinetochores to spindle microtubules and ensuring correct chromosome alignment on the metaphase plate[1][4]. It also supports DNA double-strand break repair by promoting homologous recombination, and helps maintain the expression and stability of anti-apoptotic proteins such as Mcl-1[1][3][5]. Pathogenic variants in CHAMP1 cause an autosomal dominant neurodevelopmental disorder characterized by intellectual disability, severe speech and motor delays, and dysmorphic features[2][7]. There is emerging evidence of its involvement in drug resistance mechanisms in cancer via anti-apoptotic pathways, but there are currently no approved drugs targeting CHAMP1 directly[3][5].
