CHTF18 is an evolutionarily conserved protein and an essential subunit of the Replication Factor C-like complex (CTF18-RLC). It coordinates DNA replication, repair, and cohesion establishment during cell division, safeguarding chromosomal integrity. CHTF18 is involved in ATP-dependent loading of PCNA onto DNA, modulation of DNA polymerase activity, and activation of the replication stress response. It plays key roles in chromosome transmission fidelity, especially in germline development and meiosis, and its deregulation is associated with subfertility and cancer due to genome instability. CHTF18 interacts with CTF8, DCC1, RFC2, RFC3, RFC4, RFC5, PCNA, and DNA polymerase POLH, functioning centrally in genome maintenance and chromatid cohesion during S phase and metaphase-anaphase transition.
