Overview

Chromosome X open reading frame 38 (CXorf38) encodes a 319-amino acid protein with a conserved DUF4559 domain and primarily cytosolic or nuclear localization[2]. It is classified as an "uncharacterized" protein, with limited molecular function established, but has been implicated as a prognostic marker in several cancers (notably lung adenocarcinoma, bladder, and ovarian cancers), showing variable expression patterns and methylation status in malignant disease, in association with smoking and sex-specific gene regulation[1][2][3][4]. Functional studies indicate that CXorf38 serves as a tumor suppressor in lung adenocarcinoma, where forced expression inhibits cell proliferation and colony formation, and it is downregulated in both ZIP8-knockout cells and certain cancer models[4][6]. CXorf38 escapes X-chromosome inactivation in a tissue- and population-dependent manner, with female-biased expression in some tissues[2]. It interacts physically with proteins involved in transcriptional regulation, cell growth, and developmental pathways (e.g., PAX5, PAX6, NFYC, CDHR3, MEOX2, and DDIT4L)[2]. There are currently no established drugs, mechanisms of action, or well-documented safety considerations specifically targeting CXorf38, and its precise molecular role remains to be fully characterized.
