Cilia- and flagella-associated protein 298 (CFAP298) is a cytosolic and nuclear protein encoded by the CFAP298 gene on chromosome 21q22.1. The protein is mainly involved in the assembly of axonemal dynein arms, regulating motile cilia function critical for correct ciliary movement and positioning, and thus planar cell polarity. Mutations in CFAP298 disrupt dynein arm assembly, leading to primary ciliary dyskinesia and related disorders. The protein is expressed in most tissues, except for relatively lower levels in the heart, liver, and certain brain regions, and has notable roles during early development. No small-molecule drugs or therapeutic agents are known to target CFAP298 directly. The gene’s altered expression is associated with prognosis in certain cancers (acute myeloid leukemia) and neurodegenerative disorders (Alzheimer's disease), but it is not considered a direct therapeutic target or classic receptor, enzyme, or transporter[1][2][3][5][9].
