Cilia- and flagella-associated protein 36 (CFAP36) is a structural protein localized to the ciliary transition zone and is involved in regulating ciliary transport processes. It contains a BART-like domain and directly interacts with the small GTPase Arl3, modulating its localization and potentially facilitating presentation to GTPase-activating proteins. The protein is expressed in multiple human tissues, including the brain, testis, heart, lung, pancreas, and spleen. Loss or mutation of CFAP36 is linked to disorders such as retinitis pigmentosa and Joubert syndrome, likely due to its importance in ciliary integrity and function. CFAP36 is noted as an autoantigen in some cases of paraneoplastic neurological syndromes with anti-Yo antibodies. It is not recognized as a classical drug target such as a receptor or enzyme, but rather plays a structural and regulatory role in the ciliary apparatus.
