Overview

The Ciliary neurotrophic factor receptor alpha (CNTFR) is a key component of a heterotrimeric receptor complex that mediates the biological actions of ciliary neurotrophic factor (CNTF). This complex is unique because the alpha subunit is anchored to the cell membrane by a glycosylphosphatidylinositol (GPI) linkage and lacks an intracellular signaling domain; signaling is instead initiated through the recruitment of two transmembrane proteins, gp130 and the leukemia inhibitory factor receptor (LIFR) (Davis et al., 1991, Science; UniProt P26992). Activation of this complex triggers the JAK/STAT, PI3K, and MAPK pathways, which are essential for the survival, differentiation, and maintenance of various neuronal types, including motor and sensory neurons (Ip et al., 1993, Endocrinology). Beyond its neuroprotective roles, the CNTFR complex is a significant regulator of energy metabolism in the hypothalamus, making it a target for anti-obesity drug development (Lambert et al., 2001, PNAS). Clinical efforts have focused on using CNTF analogs like Axokine to induce weight loss or using encapsulated cell technology (NT-501) to deliver CNTF for retinal diseases (Ettinger et al., 2003, JAMA; Sieving et al., 2006, PNAS). Despite its potential, therapeutic application has been hindered by the development of neutralizing antibodies and systemic inflammatory side effects.

Mechanism of action

Agonist binding to the CNTFR-alpha subunit triggers the recruitment of gp130 and LIFR-beta, forming a tripartite complex that activates the JAK/STAT, MAPK, and PI3K signaling pathways to promote neuronal survival and modulate energy homeostasis.
