Overview

Circular RNA 0058051 (circ_0058051) is a non-coding RNA molecule produced by the back-splicing of exons from the PTPN22 gene (Protein Tyrosine Phosphatase Non-Receptor Type 22) [PMID: 32856534]. It functions as a competitive endogenous RNA (ceRNA), acting as a molecular sponge for microRNAs such as miR-542-3p. By sequestering these microRNAs, circ_0058051 prevents them from binding to their target messenger RNAs, which leads to the upregulation of genes involved in tumor progression. In clinical studies, circ_0058051 has been found to be significantly overexpressed in pituitary adenoma tissues compared to normal tissues [PMID: 32856534]. Its high expression levels are associated with increased cell proliferation, migration, and invasion, as well as the inhibition of apoptosis in tumor cells. Consequently, circ_0058051 is considered a potential diagnostic and prognostic biomarker for pituitary tumors. From a therapeutic perspective, it represents a novel target for RNA-interference (RNAi) or antisense oligonucleotide (ASO) therapies aimed at silencing its oncogenic activity. Currently, there are no approved drugs or clinical-stage candidates specifically targeting circ_0058051, and research remains in the preclinical stage. Challenges for targeting this molecule include the need for efficient delivery systems to the central nervous system and the avoidance of off-target effects.
