Circular RNA 0067557 (hsa_circ_0067557) is a stable, non-coding RNA molecule derived from the back-splicing of the RBP1 gene (PeerJ, 2021). It is significantly upregulated in colorectal cancer, particularly within exosomes derived from cancer-associated fibroblasts (CAFs), where it promotes tumor progression, metastasis, and resistance to chemotherapy (BMC Cancer, 2024). Mechanistically, circ_0067557 acts as a molecular sponge for Lin28A and Lin28B and recruits the transcription factor BHLHE40 to the OTUB2 promoter, thereby activating pathways that drive epithelial-mesenchymal transition (Biology Direct, 2026). Beyond its role in oncology, it has also been identified as a component of a competitive endogenous RNA network in preeclampsia, regulating the PI3K-Akt signaling pathway (Annals of Medicine, 2021). Due to its functional importance and presence in liquid biopsy samples, circ_0067557 is being investigated as both a diagnostic biomarker and a potential therapeutic target. Currently, no clinical drugs target this molecule, but experimental research utilizes RNA interference to suppress its oncogenic activity.
