Overview

Circular RNA transcription factor Dp-2 (circTFDP2), also known as hsa_circ_0008304, is a circular RNA molecule derived from the TFDP2 gene. It has emerged as a significant regulator in cancer biology, particularly in prostate cancer, where it is frequently upregulated in tumor tissues and exosomes. circTFDP2 functions primarily by interacting with the poly(ADP-ribose) polymerase 1 (PARP1) protein. By binding to the DNA-binding domain of PARP1, circTFDP2 prevents its cleavage by caspase-3, which in turn inhibits DNA damage-induced apoptosis and promotes the proliferation and metastasis of cancer cells. Beyond its role in prostate cancer, circTFDP2 has been found to be downregulated in T-cell acute lymphoblastic leukemia, suggesting a context-dependent role in different malignancies. Due to its stability and presence in exosomes, it is considered a potential diagnostic biomarker and a promising therapeutic target for the development of RNA-based or exosome-targeted interventions.
