Overview
Cisplatin is a small molecule, platinum-based chemotherapy drug classified as an alkylating agent. It exerts its cytotoxic effects primarily through interaction with DNA, forming intra-strand and inter-strand cross-links. This disrupts DNA replication and transcription, leading to cell cycle arrest and apoptosis. Common toxicities include nephrotoxicity, ototoxicity, neurotoxicity, nausea/vomiting, and myelosuppression. Cells may develop resistance via increased DNA damage repair, reduced drug accumulation, or increased detoxification.
