Overview

CKLF-like MARVEL transmembrane domain-containing 1 (CMTM1) is a protein-coding gene in the chemokine-like factor superfamily, possessing both chemokine-like and transmembrane-4 superfamily features. It is broadly expressed in human tissues, especially in testis and various tumors. CMTM1 promotes tumor cell proliferation, survival, and migration in a range of cancer types and is overexpressed in several cancers where it correlates with poor prognosis and resistance to standard therapies. Functionally, it bridges roles in cancer biology, immune signaling, and tissue development. CMTM1 is considered a potential therapeutic target and prognostic biomarker in oncology research, though no drugs currently target it directly. The readthrough product from the CKLF-CMTM1 locus exists, but it is rarely referenced as a distinct biological or pharmacological entity; most literature focuses on the canonical CMTM1 protein.

Mechanism of action

Promotes cancer cell proliferation; Enhances tumor cell invasion/migration; Confers chemoresistance (for example, resistance to platinum-based chemotherapy and tumor necrosis factor–induced apoptosis, largely by modifying signaling and apoptosis pathways).

05

Safety considerations

As a potential cancer target, safety concerns may include unwanted immune modulation or toxicity to normal proliferative tissues, but these have not been well-characterized as there are currently no approved therapeutics.
