Overview

CKLF like MARVEL transmembrane domain-containing 7 (CMTM7) is a human protein encoded by the *CMTM7* gene on chromosome 3p22.3[1][2][3]. It belongs to the CKLF-like MARVEL transmembrane domain-containing family, which are structurally related to chemokines and the transmembrane 4 superfamily[2][3]. CMTM7 is widely expressed, especially in leukocytes, spleen, breast, and lung, with at least six splicing isoforms[3]. The predominant isoform, CMTM7-001, is the most studied. CMTM7 acts mainly as a **tumor suppressor**: its expression is typically decreased in various cancers, and its forced overexpression suppresses cancer cell proliferation and tumor formation in models[1][2][3]. Mechanistically, CMTM7 is involved in the regulation of the EGFR/PI3K/AKT pathway by promoting EGFR internalization, degradation, and downregulating downstream signaling; it also regulates cell cycle progression through effects on cyclin-dependent kinases and p27[2][3]. Its immune system roles and potential links to other disease settings are under ongoing study[3][4]. No drugs are currently known to directly interact with CMTM7, and its use as a clinical biomarker or therapeutic target remains under investigation[1][3].
