CKLF-like MARVEL transmembrane domain-containing protein 1 (CMTM1) is a member of the chemokine-like factor superfamily and the MARVEL domain-containing family of proteins, sharing structural similarities with both classical chemokines and transmembrane 4 superfamily proteins. Encoded by the CMTM1 gene on chromosome 16, the protein exists in at least 23 isoforms generated by alternative splicing. CMTM1 is weakly expressed in most normal tissues but is strongly expressed in the testes and various cancer types, where some isoforms (notably CMTM1-v17) promote cancer cell proliferation, invasion, and chemoresistance, correlating negatively with patient prognosis. In contrast, isoform CMTM1-v5 has been shown to induce apoptosis in certain lymphoma cell lines and suppress hematological tumor growth in mouse models. Accordingly, CMTM1 has emerged as both an oncogenic factor and a potential therapeutic target, as well as a prognostic biomarker in solid and hematological malignancies. Further investigation into isoform-specific functions, disease associations, and responsiveness to targeted therapies is active.

Experimental mechanisms involve: - Suppression of cancer cell proliferation and invasiveness by inhibiting CMTM1 - Induction of apoptosis in lymphoma cells by specific isoforms (CMTM1-v5) - Modulation of apoptosis and chemoresistance (CMTM1-v17’s role in resistance to chemotherapy and TNF-α–induced apoptosis)
