Overview

CKLF-like MARVEL transmembrane domain-containing protein 2 (CMTM2) is a multifunctional transmembrane protein encoded by the CMTM2 gene on chromosome 16q22.1[1][2][3]. It contains a MARVEL domain, is part of the chemokine-like factor superfamily, and localizes primarily to membranous and cytoplasmic compartments such as the plasma membrane and endoplasmic reticulum near the Golgi apparatus[2]. CMTM2 is highly expressed in normal testicular tissues, bone marrow, and various blood cells, and plays roles in steroidogenesis and immune cell regulation[1][2]. Its altered expression (including epigenetic modifications such as promoter hypermethylation) has been observed in multiple cancers and immune diseases, typically acting as a tumor suppressor by inhibiting proliferation, invasion, and metastasis; higher expression correlates with improved prognosis[1][3]. CMTM2 can serve as a biomarker for disease severity and prognosis in certain cancers, and as a marker for cutaneous T-cell lymphoma variants (e.g., Sézary syndrome)[3].
