Overview

CKLF-like MARVEL transmembrane domain-containing protein 6 (CMTM6) is a type I transmembrane protein encoded by the CMTM6 gene, located on chromosome 3p22.3 in humans[1][7]. It is a member of the CKLF-like MARVEL transmembrane domain-containing (CMTM) family, which is involved in various physiological and pathological processes including immune regulation, inflammation, cancer, cardiovascular disease, and autoimmune conditions[1][2]. CMTM6 is best known for its critical role in regulating the surface expression of immune checkpoint protein PD-L1 on tumor cells, thereby modulating anti-tumor immune responses and immune evasion in cancer[1]. The protein is also implicated in ocular inflammatory responses, improvement of epithelial barrier function, and other immunoregulatory activities[6][7]. No direct therapeutic drugs are currently approved specifically for CMTM6, but it is a candidate for intervention in cancer immunotherapy due to its impact on PD-L1 expression and immune responses[1].
