CKLF-like MARVEL transmembrane domain-containing protein 8 (CMTM8) is a four-pass transmembrane protein belonging to the CKLF-like MARVEL transmembrane domain-containing family, characterized by a MARVEL domain involved in vesicle trafficking and membrane apposition[1][3][4]. CMTM8 is broadly expressed in normal tissues and functions primarily as a tumor suppressor, with reduced expression reported in various cancers, such as non-small-cell lung, colorectal, gastric, esophageal, bladder, and brain tumors[1][2][3][4]. Functionally, CMTM8 regulates internalization and signaling of important cell-surface growth factor receptors (notably, EGFR), inhibits cancer progression by limiting proliferation, migration, and invasion of tumor cells, and promotes apoptosis through both caspase-dependent and independent mechanisms[2][3]. Currently, CMTM8 is not directly targeted by any approved therapeutics, but research suggests its expression might serve as a useful biomarker for cancer prognosis and a potential future therapeutic target[1][3][5].
Not established for approved drugs (no direct drug targeting known). Acts biologically by promoting internalization of growth factor receptors (notably EGFR) to suppress signaling. Induces apoptosis via both caspase-dependent and -independent pathways.
