The Class III phosphatidylinositol 3-kinase complex (PI3K-III complex) is a conserved multiprotein assembly responsible for the generation of phosphatidylinositol 3-phosphate (PI3P), a lipid signaling molecule that initiates autophagosome formation and regulates endosomal trafficking. The canonical mammalian complex consists of the catalytic subunit VPS34 (PIK3C3), regulatory subunit VPS15 (PIK3R4), Beclin 1, and either ATG14 or UVRAG, depending on autophagic versus endocytic function. The complex plays a central role in cellular responses to nutrient status, organelle quality control, and maintenance of protein homeostasis. Dysfunction or dysregulation of the PI3K-III complex is implicated in cancer, neurodegeneration, and cardiovascular disease. It is widely studied as a therapeutic target, although broad inhibition may pose safety risks due to its essential roles in autophagy and organ function.
