Clathrin coat assembly protein AP180 (SNAP91) is a neuronal endocytic adaptor protein essential for clathrin-mediated endocytosis, especially at synaptic membranes. It contains specialized domains, including an intrinsically disordered region that promotes membrane curvature and an ANTH domain that binds to SNARE motifs, supporting both assembly of clathrin coats and selective sorting of vesicular proteins such as synaptobrevin 2. SNAP91 ensures proper synaptic vesicle recycling; disruptions in its function lead to deficits in neurotransmission and have been linked to neurological diseases such as Alzheimer's disease and epileptic seizures. Genetic studies also suggest an association with craniofacial disorders such as cleft lip/palate, but the gene's primary expression is in the adult brain. There are currently no known approved drugs that directly interact with SNAP91, and it is not established as a therapeutic target in current clinical practice.
