Clathrin heavy chain 1 is a large protein found on the cytoplasmic face of intracellular organelles, particularly clathrin-coated vesicles and pits. These vesicles are crucial for the internalization and trafficking of receptors, endocytosis of macromolecules, and maintenance of cellular homeostasis. The basic clathrin triskelion structure is formed by three heavy chains and three light chains, assembling into lattices that curve to generate vesicle coats. Clathrin heavy chain also interacts with several adaptor proteins and is essential in autophagy and mitosis. Pathologically, chromosomal rearrangements that fuse CLTC with receptor tyrosine kinases, such as ALK, can drive constitutive kinase activation, contributing to malignancy in a subset of lymphomas and other tumors
