Clathrin-mediated endocytosis is a fundamental cellular process in eukaryotic organisms whereby specific cell-surface molecules, such as receptors and nutrients, are internalized via vesicles coated with the protein clathrin. It involves a multistep sequence including initiation at the plasma membrane (via adaptor proteins like AP-2 and lipid PIP2), clathrin coat assembly, invagination, vesicle scission (by GTPase dynamin), uncoating (by auxilin and Hsc70), and transport to early endosomes. CME plays central roles in cell signaling, nutrient uptake, synaptic function, and is exploited by pathogens for cellular entry. The pathway involves over 50 proteins, and dysfunction may contribute to cancer, neurological, and metabolic diseases.
