Overview

Claudin-11 (CLDN11) is an integral membrane protein belonging to the claudin family, which forms the backbone of tight junction strands in multiple organ systems. It is a major structural component of myelin in the central nervous system, where it is essential for the assembly of specialized paracellular barriers and the regulation of ion flow and electrical resistance, thereby enabling normal nerve conduction. In the testis, claudin-11 is indispensable for blood-testis barrier formation by Sertoli cells, crucial for spermatogenesis and immune privilege. Functional loss or mutation of CLDN11 causes syndromes such as hypomyelinating leukodystrophy (HLD22), deafness, and infertility. Claudin-11 also interacts with other proteins to regulate oligodendrocyte proliferation and migration, and emerging data suggest it may serve as an autoantigen in autoimmune demyelinating disease[1][2][3][4][5][7].
