Overview

Claudin-2 is a 230-amino acid, tetraspan transmembrane protein that forms the backbone of tight junction strands in leaky epithelial tissues. Its extracellular domains line selective paracellular pores through which small cations and water can cross, contributing to energy-efficient transport especially in the kidney proximal tubule and intestinal tract. Regulation of Claudin-2 expression and phosphorylation affects its localization, permeability, and turnover; altered expression is linked to pathological states such as cancer progression, inflammatory disease, and kidney dysfunction. Beyond simply controlling epithelial permeability, Claudin-2 participates in signaling networks that modulate cell proliferation and fate, making it a multifaceted therapeutic target.
