Claudin domain-containing protein 1 (CLDND1) is a transmembrane protein belonging to the claudin family and is involved in the formation and regulation of tight junctions in various tissues[1][5][7]. These junctions are critical structures that control paracellular barrier permeability and help maintain cell–cell adhesion, particularly in the central nervous system and endothelium[1][6]. In the brain, CLDND1 expression is important for vascular barrier integrity, with its downregulation leading to increased endothelial permeability following injury[1]. CLDND1 is dynamically regulated by physiological and environmental factors, including transcriptional control by RORα and MZF1, and post-transcriptional regulation by microRNAs[1]. In immune cells, particularly CD4+ T-cells, CLDND1 may function as a negative regulator of proliferation or induce cell death, suggesting a role in immune tolerance and possibly in autoimmune disease[3]. Its expression and regulation have been implicated in cancer cell survival, cerebrovascular disease, and immune modulation, but it is not yet considered a direct therapeutic target nor are there known drugs targeting it specifically[1][3][7].
