Overview

Cleavage and polyadenylation specificity factor 4 (CPSF4, also known as CPSF30 or NEB1) is a 30 kDa protein component of the CPSF complex, critically involved in 3'-end processing and polyadenylation of pre-mRNA[1][2][3]. It contains several zinc finger motifs and recognizes the polyadenylation signal on pre-mRNA, aiding in cleavage and polyadenylation[2]. CPSF4 regulates RNA maturation and, through these functions, impacts cell proliferation and apoptosis; its overexpression in lung adenocarcinoma is associated with poor prognosis, while its inhibition suppresses tumor growth[1]. CPSF4 interacts with the influenza virus NS1 protein, which blocks host cell mRNA processing to support viral replication[3]. Although not currently a therapeutic drug target, CPSF4 has potential as a prognostic biomarker and possible future target in oncology[1].

Mechanism of action

Not directly targeted by any approved drugs; functional inhibition (e.g., by siRNA) can reduce tumor growth and promote apoptosis[1]. The influenza A NS1 protein inhibits its function during viral infection[3].

Safety considerations

No clinical therapeutic targeting or safety data available. Biological concerns include its essential cellular role in mRNA maturation—systemic inhibition may cause broad cytotoxicity or impact normal cell function
