Cleavage and polyadenylation specificity factor subunit 5 (CPSF5, also known as NUDT21 or CFIm25) is an RNA-binding protein that functions as a central component of the cleavage factor Im (CFIm) complex, which is essential for the 3' RNA cleavage and polyadenylation processing of pre-mRNA[1][2][5][6]. It is responsible for the recognition of UGUA sequence elements in pre-mRNAs, facilitating the early steps in assembly of the 3' end processing complex and recruiting other processing factors[1][2]. CPSF5 possesses a Nudix domain but lacks hydrolase activity, distinguishing it from many Nudix family members that do catalyze nucleotide hydrolysis[2][3]. Dysfunction of CPSF5 impacts alternative polyadenylation, a process linked to cell differentiation, neural function, and cancer development[3][7]. There are no well-characterized small molecule drugs or approved therapeutic interventions targeting this protein, but its role in mRNA processing makes it a potential target in research, particularly in cancer and RNA dysregulation disorders.
