Overview

Cleft lip and palate transmembrane protein 1-like protein (CLPTM1L), also known as CRR9, is a multi-pass transmembrane protein that plays a critical role in cell survival and resistance to apoptosis (UniProt Q96KA5). It is frequently overexpressed in various malignancies, including lung, pancreatic, and ovarian cancers, where it contributes to tumor progression and resistance to DNA-damaging agents like cisplatin (PubMed: 21602877). CLPTM1L has been shown to interact with the catalytic subunit of telomerase (TERT) and is involved in the regulation of telomere maintenance and the DNA damage response (PubMed: 24501121). Genetic variations in the CLPTM1L gene, particularly the SNP rs401681, are strongly associated with an increased risk of developing lung cancer and other solid tumors (PubMed: 18392013). Because its inhibition sensitizes cancer cells to chemotherapy and induces apoptosis, CLPTM1L is considered a promising therapeutic target for the development of novel anti-cancer strategies (PubMed: 25605116). Current research focuses on the development of small molecule inhibitors and targeted antibodies to disrupt its pro-survival signaling in tumor cells.
