Overview

CLOCK-interacting pacemaker (CIPC) is a protein-coding gene that encodes a nuclear protein involved in the negative regulation of circadian transcriptional activity as part of the mammalian circadian clock. CIPC has been identified as a negative-feedback regulator that can inhibit the activity of the CLOCK–BMAL1 transcriptional complex, fine-tuning circadian rhythms. Although it was initially thought to be essential for circadian rhythm regulation, genetic disruption in mice showed no major alterations in core clock gene expression except for a minor effect on Per1, suggesting CIPC is dispensable for core circadian oscillator function but may modulate its precision. CIPC also contributes to cell cycle control, suppressing cell proliferation and affecting metabolic pathways via subcellular localization. There are currently no therapeutic drugs targeting CIPC, and its involvement in disease is limited to basic regulatory and metabolic roles rather than as a primary disease gene[1][3][5].
